shows an example of a typical ICa recorded by scientists at
OCP recorded under our
standard conditions for
the measurement of ICa in single
left ventricular myocytes using
the whole-cell patch-clamp technique.
The time course for the effect of nifedipine is shown on the left
panel and the bar above the records denotes application of
nifedipine. Representative calcium current records (taken from the
same single cell) recorded in the absence (control, black) and
presence of nifedipine (red and denoted by arrow) is shown in the right panel.
Nifedipine (300 nM, application denoted by the bar,
note the dotted line denotes zero current level) caused complete
block of ICa.
Nifedipine inhibited ICa
with an IC50 of 5.4 nM.
